Football rules, help?

I just started getting into American football and am just starting to learn the rules so I may come across as dumb here buy give me a break I'm new to the game lol. I'm from Scotland so I'm used to watching football (soccer) and you are only allowed three substitutions in the game unless its a friendly and then you are allowed up to six. I know in American football that you are allowed unlimited substitutions and that the whole team can be substituted due to the fact that there will be players that are good at offensive play and one's that are good at defensive play (I think, could be wrong lol)
BUT I was wondering since this can happen are you allowed to substitute a player off and then substitue him back on?
Also another thing I have been pondering is: Will there be a quarterback on the defensive's team or when a team is defending will they not have a quarterback? Don't know why i think they wouldn't I just wondered lol
Please help and sorry if i got a lot wrong but i literally just started watching last week :) and I really enjoy it but i think i would like it better if i knew what these rules were :)
Thanks, I thought the quarterback was just for offensive play but like i said i'm new and wasn't sure :)
