Hop on and off bus tour London - is there one with a real tour guide on board...

...or are they all just recordings? Also, if I get on one of these tours from say, the London Eye (or closest stop), how much time should I allow to get to Madame Toussards? I'm trying to work out what time I should fast track my entry ticket into Mme Toussards and still utilise the hop on and off bus tour. Many thanks if you can help me.
