Fellow lesbians say they often can tell a girl is lesbian by the eye contact she gives them and often by the way they are when they speak to you.

Also, how would this change if the lesbian were attracted to you? Consider both lesbians who are out and lesbians who are in the closet.

It has more to do with being perceptive of peoples' body language, regardless of orientation. Lesbians don't necessarily respond differently to a greeting when compared to a straight girl. More often than not, it's a gut feeling that LGBT's get---and that comes with time and learning to "read" people based on subtle (and sometimes, not so subtle) cues.

That said, nothing is 100 percent.
