Do you meen combine at the same time, or just have the humor, drama, and suspense as separate parts in the same anime? It depends on the anime, but both are possible if the anime staff know what they're doing and pace everything accordingly.

I think a good example of this is fate/zero. There's one Master/Servant pair, Waver Velvet and Rider, that act as the comedy relief duo in a series that would otherwise be entirely serious. They did a great job with writing them in. Another good example is CLANNAD, which has a lot of lighthearted funny moments but it's precisely because of those moments that the tearjerking scenes hit so hard.

A show that did this badly was Angel Beats!. I think it was simply because it was too rushed, and the transition was so abrupt in each episode that it felt like the anime was having a mood whiplash at times.
