Which is the positive or negative lead on my speaker wires?

hahah it matters a lot. Speakers won't work if you mix up the wires. Typically the positive is a red wire and the negative is black. Sometimes the positive will be white as well.
 
I cut out my rear speakers from my car to replace them. But i dont know how to determine the (+) or (-) wires.
1. There are no symbols on original speakers to help determine
2. The colorcodes are different colors than the wires that end to the head unit.

Is it possible to use a multimeter to see what lead is pos or neg? if so how..
