If you only use two channels on a 4 channel amplifier, would the component speaker

still receive full output? In my case, I've go a 10-inch, single voice coil, 4 ohm 500max/120rms sub hooked up to two terminals (one positive and one negative) on a 4 channel amplifier rated through the following set ups (which are vaguely understood):
80W x 4 ch @ 2 ohm = 320W
65W x 4 ch @ 4 ohm = 260W
160W x 2 ch bridged

The other two terminals (another positive and negative) are not in use.

I'm asking this because I'm looking to replace the one mentioned above with one of the following:

...a 10" 4-ohm, 1000max/200rms sub

or

...a 12" 4-ohm, 1200max/250rms sub

If I stayed with the same wiring setup, and considering whether or not the amp is pumping optimal power based on the wiring, which would be a better buy?

And if you did recommend to upgrade the 10 inch sub with another--albeit more powerful--10 inch sub, should I get a bigger box or could I simply punch holes into the original closed box to accommodate the higher rated boom?
